چکیده
This application note demonstrates the utility of the DOSY package in the VnmrJ 3.1 software for both 2D and 3D HR-DOSY analysis of complex mixtures, by first elucidating the presence of two components in an epoxy mixture (2D), and then proving that a minority component in a protein mixture was in fact a smaller derivative of the major component (3D).

High Resolution Diffusion-Ordered Spectroscopy (HR-DOSY)
This application note demonstrates the utility of the DOSY package in the VnmrJ 3.1 software for both 2D and 3D HR-DOSY analysis of complex mixtures, by first elucidating the presence of two components in an epoxy mixture (2D), and then proving that a minority component in a protein mixture was in fact a smaller derivative of the major component (3D).
متن کامل3D DOSY-TROSY to determine the translational diffusion coefficient of large protein complexes.
The translational diffusion coefficient is a sensitive parameter to probe conformational changes in proteins and protein-protein interactions. Pulsed-field gradient NMR spectroscopy allows one to measure the translational diffusion with high accuracy. Two-dimensional (2D) heteronuclear NMR spectroscopy combined with diffusion-ordered spectroscopy (DOSY) provides improved resolution and therefor...
متن کاملMatrix-assisted diffusion-ordered spectroscopy: mixture resolution by NMR using SDS micelles.
Diffusion-ordered spectroscopy (DOSY) is a powerful technique for mixture analysis, but in its basic form it cannot separate the component spectra for species with very similar diffusion coefficients. It has been recently demonstrated that the component spectra of a mixture of isomers with nearly identical diffusion coefficients (the three dihydroxybenzenes) can be resolved using matrix-assiste...
متن کاملDirectly Decoupled Diffusion-Ordered NMR Spectroscopy for the Analysis of Compound Mixtures
For the analysis of compound mixtures by NMR spectroscopy, it is important to assign the different peaks to the individual constituents. Diffusion-ordered spectroscopy (DOSY) is often used for the separation of signals based on their self-diffusion coefficient. However, this method often fails in the case of signal overlap, which is a particular problem for (1)H-detected DOSY spectra. Herein, a...
متن کاملDiffusion-Ordered (DOSY) NMR of Solute Exchange Across the Human Erythrocyte Membrane: including an application of Post- Widder Laplace inversion
Nuclear magnetic resonance (NMR) spectroscopy was used to study exchange of three distinctly different molecules across the membranes of human red blood cells (RBCs). In studying water, t-butanol, and dimethyl sulfoxide we exploited the marked differences in the apparent diffusion coefficients of these species inside and outside the cells in suspensions. The measurements were made with diffusio...
